Tec-induced NFAT activation requires intact PH and SH2 domains. Jurkat T cells were transfected with an NFAT-luciferase reporter construct and 5 μg of wild-type HA-Tec or constructs with inactivating point mutations of the PH domain, the proline-rich region, or the SH3, SH2, or kinase domain.
