(A) Monocytes were pre-incubated for 30 min with 50 µM Ac-YVAD-cmk, or left untreated, before being exposed for 6 h to 40 nM αSyn F or vehicle. IL-1β released into the culture supernatants was evaluated both by ELISA and immunoblot analysis. The same supernatants were assessed for the accumulation of active caspase-1 by immunoblot. Results are the mean ± S.D. of 3 experiments conducted in duplicate with cell preparations obtained from 3 different donors. The immunoblot is from one representative donor. ***p<0.001 (B) Real-time PCR of NLRP1 and NLRP3 in monocytes stimulated for 2, 6 and 18 h with αSyn F.
