Inhibition of NAMPT resulted in attenuation of serine biosynthesis in cancer cells. A2780 (A) and HCT116 (B) cells were grown and treated in duplicate with FK866 at various concentrations and ± nicotinic acid (10 μm) as described (“Experimental Procedures”). After the treatment, cells were collected and processed for the analysis of phosphoserine (p-Serine) and α-ketoglutarate (α-KG) levels by LC-MS (“Experimental Procedures”).
