Decreased CrAT activity disrupts mitochondrial bioenergetics and ATP generation in pulmonary arterial endothelial cells. PAEC were transiently transfected with a scrambled siRNA or a specific CrAT siRNA for 48 h and the effect on mitochondrial respiration determined using the Seahorse XF24 analyzer. In the CrAT siRNA transfected cells there was a significant decrease in the (A) amount of oxygen consumed (OCR) for ATP production and (B) this corresponded with a reduction in cellular ATP levels.
