Assay conditions: 100 mM Tris-HCl (pH7.8), 30°C, 20 mM NaHCO₃, 10 μCi/mL NaH¹⁴CO₃, 6 mM MgCl₂, 1 mM ATP, 10 mM biotin, saturating concentrations of acetyl CoA (see Materials and Methods). The reported errors are the standard errors of the estimates of the k_cat from the linear regression analysis of the time-courses of carboxybiotin formation.
