For each assay, 2.0 μM Met471Cys TβM was incubated with buffer (100 mM KPi, pH 6.0), KCl (100 mM), ascorbate (50 mM), catalase (100 μg/mL), CuSO4 (10 μM). Following the addition of 1 mM tyramine, aliquots (115 μL) were removed at 5, 10, 15, 30, 60, 120, 240 and 360 min intervals, quenched with 1.5 μL of 70 % HClO4, and assayed for octopamine formation. Data are shown after 1, 4, and 6 h.
