(A) Splenocytes of WT and RAG−/− mice were electronically gated as CD11chiCD11b+/−CD8α+/−B220− population analyzed for expression of the indicated markers. Data are representative of at least four or five mice per group in six independent experiments. (B) Splenic cDCs isolated from WT or RAG−/− mice were loaded with OVA257–264 peptide or OVA protein for 1 h. CFSE-labeled OT I T cells were incubated for 1.5 d (peptide) or 2 d (OVA) with cDCs at a 10:1 ratio. The proliferative response of T cells was enumerated by flow cytometry. Numbers of proliferating cells are shown (mean + SEM). Data are representative of multiple independent experiments with a minimum three mice per group.
