Analysis of the binding of the survivin-targeting compound to the protein. (a) Binding of S12 to purified recombinant survivin proteins was tested by ITC. Single-site point mutagenesis was performed to create the survivin mutants F86A and V89Y. The native and the mutant proteins were produced and purified from bacteria. Note that the two mutant forms of survivin did not bind to S12 in ITC assays. (b) Circular dichroism spectra of native and mutant survivin proteins. Mut86: Sur(F86A); Mut89: Sur(V89Y).
