VEGF-A expression in human pancreatic cancer is correlated with the sensitivity of cancer cells to VV in hypoxia and normoxia. (A) VEGF expression in four pancreatic cell lines. MiaPaca2, CFPac1, Panc1, and SUIT-2 cells were seeded in 6-well plates and cultured under normoxic (20% O2) or hypoxic (1% O2) conditions for 48 h until cells reached 70% confluence. The supernatant was removed, and VEGF-A protein levels were quantified using ELISA. Experiments were performed in duplicate, and results represent the data from three separate experiments. An unpaired Student t test was performed to compare the mean VEGF-A levels. *, P < 0.05. (B) VEGF expression and viral replication in human pancreatic cancer cell lines. MiaPaca2, CFPac1, Panc1, PaTu8988t, PaTu8988s, and SUIT-2 cells were seeded in 6-well plates and cultured under normoxic conditions until the cells were all approximately 70% confluent. Fresh supernatant was taken and used for a subsequent ELISA. The viral replication of VV in these cells (infected at an MOI of 1 PFU/cell) was measured by TCID50 assay. There was a Spearman's rank correlation (R2) of 0.77 (P = 0.02).
