PIV genomes efficiently replicate and produce VLPs upon delivery into cells in DNA form. (A) Schematic representations of plasmids encoding PIV genomes and VEEV structural proteins under the control of the CMV promoter. (B) BHK-21 cells were either infected with VEEV/GFP or particles containing the packaged VEEV/CRK4/GFP genome at an MOI of 20 infectious units/cell or transfected with plasmids carrying the indicated PIV genomes under the control of the CMV promoter. Media and cells were harvested at 24 h postinfection or -transfection, and viruses or VLPs were pelleted from equal volumes of medium and analyzed by SDS-PAGE followed by Coomassie blue staining or Western blotting using VEEV-specific antibodies.
