(a) Wild-type LNK is 575 amino acids and contains an N-terminal proline-rich dimerization domain (Pro/DD), a pleckstrin homology (PH) domain, a Src homology 2 (SH2) domain, and a C-terminal conserved tyrosine residue (Y). DEL is a five-base-pair deletion and missense mutation that leads to a frameshift and premature stop codon. E208Q is a point mutation at the beginning of the PH domain.
