Effects of AUF1 and AGO2 abundance on mRNA binding. K562 cells were transiently transfected with plasmids encoding control shRNA or shRNA against either AUF1 or AGO2. Two days after transfection of shRNA-expressing plasmids, lysates were prepared and fractionated by mRNP-immunoprecipitation with control antibodies or anti-AUF1 antibody (for AGO2-knockdown cells) or anti-AGO2 antibody (for AUF1-knockdown cells). Both input and immunoprecipitated materials were analysed by qRT–PCR, and abundances of the indicated mRNAs were plotted as a percent of the input mRNA level.
