Effects of mutations in L1 (L46A) and/or L2 (L328A) on subcellular localization of MRTF-B. COS-7 cells were transfected with expression plasmids for the indicated FLAG-tagged MRTF-B and cultured under the same conditions as described in the legend for Fig. 4. For the last 2 h, the cells were incubated with vehicle or LMB (5 ng/ml). The cells were then stained with anti-DYKDDDDK (FLAG) antibody and Hoechst 33258. The images were quantified as described under “Experimental Procedures.” N, nuclear accumulation; NC, diffuse distribution in the nucleus and cytoplasm; C, cytoplasmic localization.
