(A) IVP assay was carried out as described in materials and methods. The data represent mean ± SEM of 3 independent experiments and p-values were calculated by student t test. *p<0.05 and ns, p>0.05. (B) TBM-1 cells were transfected with either pcDNA3.1(+) vector alone or GPR158-pcDNA3.1(+) expression plasmid using Lipofectamine LTX reagent. The cell lysates were prepared using RIPA buffer at 3-days post-transfection and the western blotting for the detection of GPR158, ZO-1, occludin and N-cadherin was performed using appropriate antibodies. The same membrane was striped and reprobed for β-actin for loading control. The data represent two independent transfection experiments performed in triplicate.
