IL-10 levels in culture supernatants were quantified by ELISA after 24 h of treatment. (A) NDP-α-MSH induced IL-10 release from microglial cells. The stimulatory effect of NDP-α-MSH is not seen in the presence of the MC4R antagonist HS024 (B) or of the PPAR-γ antagonist GW9662 (C). (D) GW9662 has an inhibitory effect per se on IL-10 release from microglia. Data were analyzed by one way ANOVA followed by Dunnett’s or Bonferroni’s post test and are expressed as mean ± SEM. ***p<0.001, **p<0.01, *p<0.05 vs. control group, p<0.05 vs. NDP-α-MSH.
