Tob1 inhibits proliferation of gastric cancer cells. (A) Protein lysates (30 μg) were separated by SDS-PAGE and immunoblotted to detect the expression of Tob1 in AGS, MKN1 and MKN28 gastric cancer cells. (B) MKN28 or AGS cells were transiently transfected with myc-Tob1 (2 or 4 μg) or empty vector (4 μg) for 24 h, and cell viability was evaluated using WST assay as described in the Materials and methods section. Western blot analysis (top panel) confirmed the expression of myc-Tob1. Data are representative of three independent experiments showing a similar pattern. (C) MKN1 and AGS cells were transfected with either control si-RNA (50 nM) or Tob1 siRNA (si-Tob1, 25 or 50 nM). Cell viability was assessed after 48 h of transfection. Western blot analysis (top panel) confirmed the downregulation of Tob1.
