(A – C) TSLP enhanced Th9 effector function in vivo. Naïve CD4+ T cells from DO11.10 mice were differentiated under Th2 and Th9 skewed conditions with OVA323–339 peptide and APC. Differentiated Th cells were injected to Tslpr−/− recipients via tail vein, with PBS injection as control. Mice were analyzed after challenge with OVA or OVA+TSLP for 5 days for (A) BAL total cell and eosinophil counts, (B) BAL cytokine concentration using ELISA, and (C) tissue pathology using hematoxylin and eosin (H&E) and Periodic acid-Schiff (PAS) staining of paraffin-embedded lung sections.
