Glycosylated E2 inhibits Gag targeting to the plasma membrane. HeLa cells were transfected with Gag-Pol along with empty vector, E2, or immunoglobulin G (IgG)–E2. Twenty-three hours after transfection, cells were fixed, permeabilized, and immunostained, as described in Materials and Methods. Gag and E2 proteins were detected with anti-p17 polyclonal antisera and anti-cMyc, respectively. Gag is shown in red (center), and E2 proteins are shown in green (left). Fifty cells were counted in each group. The percentage of cells showing the presented pattern was calculated and is shown in the far right of the figure.
