YeaJ possesses DGC activity shown by c-di-GMP levels in S. Typhimurium. WT, JA002 (ΔyeaJ), and WT+pYeaJ strains of S. Typhimurium were grown on M9 medium supplemented with 0.4% glucose to mid-log phase and were collected by centrifugation. Nucleotides were extracted from the cell pellets as described in Materials and Methods and then were subjected to UPLC-MS-MS. Levels of c-di-GMP were normalized relative to the WT strain (set to a value of 1). Statistical significance was determined by a one-way ANOVA, followed by Bonferroni's correction, compared to the control (WT): ***, P < 0.001; ns, not significant.
