Mutant MrfA proteins that contain R to A substitutions in two residues essential for DNA binding by Ndt80 (Fig. 2C section a) were expressed as HIS fusions in E. coli and assayed for binding to the 39-mer. The fusion protein amounts were normalized by parallel gel electrophoretic analyses, followed by staining, and are expressed as the number of ug used in each reaction.
