Inhibition of SIRT1 and SIRT2 in MDA-MB-231 cells causes a decrease in aromatase transcript and protein. (A) MDA-MB-231 cells were treated with 100μM cambinol or inhibitor VII for 32 hours, and RNA was isolated and reverse transcribed. Quantitative PCR was performed to determine relative changes in mRNA transcripts. (B and C) MDA-MB-231 cells were treated with varying amounts of cambinol over a (B) 32-hour or (C) 24-hour period with DMSO used as a vehicle control. RNA was isolated, and cDNA was generated. PCR was performed using intron-spanning primers. For Western blot analysis, equal protein was analyzed using antibodies against aromatase and β-actin.
