Representative western blots and quantification of insulin receptor-β (IR-β) Tyr¹³⁶¹ phosphorylation and total insulin receptor (a), Ser³⁰⁷ and Tyr⁶¹² IRS-1 phosphorylation and total IRS-1 (b), phosphorylation (Ser⁴⁷³ and Thr³⁰⁸) and total Akt (c), and insulin-stimulated glucose clearance (d) in TA muscle from C57Bl/6 mice 7 days following electroporation of an empty vector (C; CON; grey bars) or CA-Jnk (T; Test; black bars) plasmid. Ser³⁰⁷ phosphorylation measurements were made in the absence of insulin stimulation; all other measurements were insulin stimulated. For signalling, insulin (1 U/kg) was injected into the hepatic portal vein and tissue was excised after 10 min. For glucose uptake, 0.5 U/kg insulin and 370,000 Bq³ 2-deoxy[³H]glucose were injected.
