Cells (2x105) were incubated with different concentrations of BaV or RPMI (control) for 60min at 37°C under 5% (v/v) CO2, after which cytotoxicity was assessed by Trypan blue exclusion. Cells were harvested 96hr after i.p. injection of thioglycollate and were incubated with BaV or RPMI (control) for 60min before addition of opsonized zymosan particles. Values represent the mean ±SEM from four animals. *p<0.05 compared to controls (ANOVA). B. Effect of BaV on superoxide anion production by migrated leukocytes. The cells were collected from mice peritoneal cavity 6hr after BaV (0.2mg/kg) or sterile saline alone (0.15M NaCl) (control) injection. 2x105cells/ml were incubated for 40min with NBT. Cells were then stained with safranin and centrifugated (400xg/5min) in cytospin. NBT positive cells were evaluated by cell count crystals that formed inside of the cell by phase contrast microscope.
