The supernatants of rat splenocytes stimulated with anti-CD3 antibody and IL-12 in the presence (super1) and absence (super2) of sinomenine (1 mmol/L) were collected and then used to induce iNOS production by primary astrocytes. A: RT-PCR was performed with the cellular RNA extracts to determine the expression levels of iNOS mRNA. B: Western blotting was performed with the cellular protein extracts to assay the level of iNOS protein. The results shown are mean±SD for three independent experiments. Significant difference between the effects of supernatants of splenocytes cultured with anti-CD3 antibody and IL-12 and simultaneously with anti-CD3 antibody, IL-12 and sinomenine on iNOS production by primary astrocytes was analyzed.*P < 0.05.
