Bacterially expressed, affinity-purified His-Bsn11 was phosphorylated in vitro by RSK 1, 2, 3 and 4 using ascending amounts (10-40 mU) of active purified kinase. Equal loading of His-Bsn11 in the assay was controlled by immunodetection using α-His in all samples. Phosphorylation of His-Bsn11 was monitored by immunodetection with α-pS2845 Bsn, which revealed increasing phosphorylation depending on the increasing concentrations of kinases. The images shown here are representative for three independently performed experiments. The bars and numbers on the left side of the blots show the sizes and positions of the molecular markers.
