DN3 were cultured on plate-bound recombinant DL4 in the presence or absence of CXCL12 (10 nM) and CHIR99021 (1, 3 or 10 µM). After 72 hours cells were harvested and analyzed for the percentages of CD4⁺CD8⁺, CD4⁺CD8⁻, CD4⁻CD8⁺ and CD4⁻CD8⁻ cells (a) and the total number of cells relative to input (b). The graph shows the mean and standard deviation of three biological replicates conducted on three different days. The p values were determined using the paired Student's t test (*  =  p<0.05, ** =  p<0.01 and *** =  p<0.001).
