DN3a were cultured on OP9-DL1 cells in the absence or presence of CHIR99021 (1, 3 or 10 µM). Cells were harvested and stained with either rat IgG2a (shown in red) or rat anti-mouse CD127 (IgG2a; shown in turquoise). Cell surface expression of CD127 on DN3 cells (CD25⁺CD98⁺) at 48 hours was analysed. The median values given for the surface expression (a) and the the graph shows the mean and standard deviation of three biological replicates conducted on three different days. The p values were determined using the paired Student's t test.
