Ag-stimulated cell ruffling and spreading are inhibited by PAO and quercetin. (A) IgE-sensitized cells were pretreated with or without quercetin (20 µM) or PAO (2 µM) for 2 minutes and then stimulated with Ag (DNP-BSA, 0.5 µg/ml) for 10 minutes. Cells were then fixed and labeled for F-actin with A568-phalloidin for analysis by confocal microscopy. Cells were scored for ruffling (B) and cell area for spreading (C) for >50 cells for each condition in three independent experiments (error bars represent S.D.). The difference between the cell area among Ag-stimulated cells with or without inhibitors was statistically analyzed using Student’s t test and was considered significant when **P < 0.01.
