Caspase inhibitors that kill microglia are able to prevent LPS-induced neuronal death in mixed neuronal/glial cultures over a 72-h time period. A–C, cultures were treated with 100 ng/ml for 48 h prior to addition of 50 μm Z-VAD (A), IETD (B), or DEVD (C) for a further 24 h prior to quantification of neuronal survival and death. Microglial survival from these experiments is presented in Fig. 1, B--D. Data represent mean values, and error bars are the S.E. of neuronal survival (n = 3). D and E, cultures were treated as described above, but where indicated, 30 μm necrostatin-1 was added 1 h prior to Z-VAD.
