Effect of FASN overexpression on drug-induced apoptosis. Stable MCF7 cells with FASN overexpression (FASN1 and FASN2) and vector-transfected control (Vec) cells were treated with 1 μM doxorubicin (DOX) or DMSO vehicle control for 48 h followed by analysis of apoptosis using DNA fragmentation ELISA (A), Western blot analysis of cleaved PARP (B), and activated caspase 8 (C8) (C). D: Western blot analysis of cleaved PARP and activated caspase 8 following a 48 h doxorubicin (0.2 µM) or DMSO treatment of MDA-MB-468 cell line transfected with FASN siRNA (Si) or scrambled (Scr) control siRNAs. Enrichment factor = ratio of DNA fragmentation of cells treated with doxorubicin/DNA fragmentation of cells treated without doxorubicin. GADPH or actin was used as a loading control for Western blot analysis. *P < 0.05.
