Basal and doxorubicin-induced levels of total ceramide in stable MCF7 cells with FASN overexpression (FASN1 and FASN2) and vector-transfected control (Vec) cells were determined using mass spectrometry as described in Materials and Methods. C: Doxorubicin-induced ceramide increase. Total ceramides were determined in stable MCF7 cells with FASN overexpression (FASN1 and FASN2) and vector-transfected control cells (Vec) following treatment with 1 µM doxorubicin for 48 h. DOX-induced ceramide level increase (fold) = total ceramide in cells treated with doxorubicin/total ceramide in cells treated without doxorubicin.
