Sucrose density gradients were performed to isolate lipid raft microdomains from the phospholipid membrane and to visualize whether Ply localized with the lipid raft fractions. Gradient fractions were blotted onto a nitrocellulose membrane, and Ply or CTxβ were detected by chemiluminescence. Fractions are numbered from least dense to most dense.
