A, marker for necrosis (propidium iodide staining) and apoptosis (YO-PRO-1) were used to stain control cells and cells overexpressing or down-regulating TRPC1 protein treated with MPP+. Rhodamine-conjugated propidium iodide and fluorescein isothiocyanate-conjugated YO-PRO-1 was added to cells treated with MPP+ for 12 h. Fluorescence images were taken immediately using either a 10× or 40× objective, and the red and green cells were counted along with total number of cells. The red box shows an enlarged image of fluorescein isothiocyanate-conjugated YO-PRO-1 on MPP+-treated cells using 100× objective.
