Specific signaling through the P2X7R induces innate Th17-biasing cytokines. Bar graphs demonstrate the relative fold change in mRNA expression of IL-1β and IL-6 24 h following cutaneous injections with 350 µM of BzATP, 1 µM KN-62 (P2X7R antagonist), BzATP + KN-62 (control), or PBS (control; gray bar). Fold-change was determined by relative qRT-PCR using the 2−ΔΔCt method in which samples were normalized to GAPDH followed by PBS controls. Data expressed as mean ± SD of triplicates. Asterisk indicates a significant difference compared to indicated treatment group, p < 0.05. Representative of three independent experiments.
