Ligand-induced assembly of the repression complex is mediated by looping of the intragenic regulatory element to the TSS of the GR gene. A chromatin capture assay was performed after digesting fixed chromatin from vehicle- or dexamethasone-treated cells with the BglII restriction enzyme. Primers flanking either side of the putative interaction sites (restriction sites) were used to amplify a specific hybrid product by RT-PCR after ligation.
