Expression of HA epitope-tagged dSLBP proteins. (A) Schematic representation of the SLBP protein encoded by the transgenes, with the RNA binding domain indicated by the shaded area. Two tandem HA tags were engineered at the start of translation. The location of the T120A and T230A mutations and the short highly conserved stretch in which they reside are indicated. (B) Western blot analysis of HA-SLBP expression in embryos. Proteins from 20 embryos were resolved on SDS-polyacrylamide gels, transferred to nitrocellulose, and HA-dSLBP detected with an anti-HA antibody. The blot was reprobed with anti-β-tubulin antibody (bottom). Lane 1, extract from wild-type control embryos lacking a transgene.
