Figure 4. — Purification of the Tim9/Tim10 complex. (A) The Tim9/Tim10 complex was isolated from mitochondria of a Neurospora strain carrying a Tim10 protein with a histidinyl tag. The mitochondria were lysed with digitonin and subjected to Ni-NTA chromatography. Fractions 5-8 from the Ni-NTA column were combined and loaded onto a Q-Sepharose ion-exchange column. Column fractions were analyzed by SDS-PAGE, and proteins were stained with Coomassie Blue. Tim 9, Tim10, and Tim10_His are indicated. (B) The proteins in fraction 3 from the ion-exchange column were analyzed by SDS-PAGE and immunodecoration with antibodies against Tim9 or Tim10. Fl. Thr., flow-through.
