SDF-1α protects myocytes from hypoxia/reoxygenation-induced cell death. Cultured myocytes were treated with 25 nmol/L SDF-1α or vehicle before exposure to 1 hour of hypoxia followed by 18 hours of reoxygenation. Myocytes also were treated with 40 μmol/L AMD3100 for 4 hours before the addition of SDF-1α. Cell death was assessed by trypan blue exclusion and LDH release. A, Trypan blue exclusion assay demonstrated increased cell death in hypoxia, which was prevented by SDF-1α (25 nmol/L); AMD3100 blocked the effects of SDF-1α.
