Tie2YIA/LAS cannot form ligand-independent dimers but is dimerized and phosphorylated upon stimulation with Ang1.
A, dimerization and localization of Tie2 were observed in Tie2-VN- and Tie2-VC-coexpressing NIH3T3 cells in the presence or absence of Ang1. Wild-type Tie2 can form dimers irrespective of Ang1 stimulation, as confirmed by BiFC. However, this dimerized Tie2 forms cluster-like aggregations and is internalized upon stimulation with Ang1. B, similar to A, dimerization and localization of Tie2YIA/LAS (Tie2LAS) is observed in Tie2YIA/LAS-VN- and Tie2YIA/LAS-VC-coexpressing NIH3T3 cells.
