Bryo targets PKCα to the cell periphery and subsequently promotes temperature-dependent perinuclear accumulation of the enzyme. IEC-18 cells on glass coverslips were treated as follows: panel a, DMSO (Control) for 1 h at 37 °C; panel b, Bryo (100 nm) for 1 h at 37 °C; panel c, Bryo for 30 min at 37 °C followed by 2 h at 4 °C; panel d, DMSO for 2 h at 4 °C; panel e, Bryo for 2 h at 4 °C; panel f, Bryo for 2 h at 4 °C followed by 1 h at 37 °C. Cells were then fixed and processed for PKCα immunofluorescence. Arrows indicate perinuclear staining, and arrowheads point to membrane staining. Magnification bars, 10 μm.
