JEV 2′-O MTase mutant shows enhanced sensitivity to IFN and IFITs. (A and B) Groups of 3-week-old 129 mice (A) and A129 mice (B) were i.p. injected with 5 × 10⁷ PFU of WT or E218A virus. The mortality was monitored for 15 days or till death. BHK-21 cells in a 96-well plate were infected with the WT or E218A mutant virus at an MOI of 5 for 1 h, followed by treatment with various doses of IFN-α A/D (10, 100, or 500 U/ml). (C and D) MTS assay (C) and plaque assay (D) were performed 48 h postinfection. (E) HEK293 cells were transfected with equal amount of plasmids IFIT-1, -2, -3, and -5 using Lipo2000 followed by infection with WT and E218A viruses at an MOI of 1. The viral titers in the supernatant were then assayed at the indicated times postinfection by plaque assay.
