Subcellular localization of NS5A. (A) Cells harboring the subgenomic replicon I389-neo/NS3-3′/5.1 were analyzed by confocal microscopy using anti-NS5A antibody in conjunction with anti-rabbit FITC-conjugated secondary antibody (green). Nuclei were stained with To-Pro-3 (red). Three-dimensional representations were reconstructed from image z-stacks. Scale bar, 10 μm. *, non-replicon-harboring cells. The yellow arrow shows where the 3D reconstructed image was cropped to display the cell sections. The yellow pictogram indicates the direction of sight. (B) Schematic representation of lentivirus vectors carrying NS5A-v1 and -v5 and β-galactosidase. Filled triangles indicate the presence of cell culture adaptive mutation S2204I. (C) Confocal analysis of cells transduced with Lenti-NS5A-v1 or -v5 was carried out as described for panel A. Scale bar, 10 μm. *, nontransduced cells. The yellow arrow shows where the 3D reconstructed image was cropped to display the cell sections. The yellow pictogram indicates the direction of sight.
