pCpG+ activates TLR9, whereas pCpG− does not. HEK 293 cells were transfected with (a) a TLR9 expression plasmid and an NF-κB responsive SEAP reporter plasmid (pNIFTY) or (b) a noncoding plasmid (pCpG+) and pNIFTY. Up to 300 µg/ml pCpG+ (92 nM plasmid, 71 µM CpG) and pCpG− (112 nM plasmid, 0 CpG), DNAse digested plasmid, or media were added in triplicate 8 h after transfection. Twenty-four post-transfection cell lysate luciferase was assayed. The difference between background activities (compare media lane between panels) is attributed to transfection variability.
