Scatter plot (A) and limiting dilution plot (B) were shown. Various doses of male LSK+CD34 HSCs (15, 45, and 150 cells/recipient mouse) were mixed with 1.5×105 WT female FVB BM cells and transplanted into lethally irradiated female FVB mice. Male donor engraftment in peripheral blood was estimated at 4 months post transplant. Poisson statistical analysis of limiting-dilution was performed to allow estimation of CRUs in each group (n = 28 for WT and n= 20 for Pim1−/− mice). The plot shows the percentage of recipient mice containing less than 1% donor male cells in the peripheral blood at 4 months after transplantation versus the number of cells injected per mouse. The horizontal line indicates the point at which 37% of the transplanted mice are non-engrafted in each group; the CRU frequency is estimated at the point where 37% of the mice are non-engrafted, by conventional Poisson statistical methods and limiting-dilution analysis. The vertical dashed lines highlight the CRU frequencies in Pim1−/− and WT mice.
