Upon ligand binding Dectin-1 is Tyr phosphorylated within the ITAM-like motif leading to Syk binding and activation. At present it is unclear whether two phosphorylated Tyr resides (i.e. dimerization of Dectin-1) are required for Syk activation since this kinase normally requires two phospho-Tyr for binding. Syk activation leads to a signaling cascade potentially involving PLC-gamma and PKC-mediated CARD9 activation. Phosphorylated CARD9 recruits and activates Bcl-10 and Malt1 leading to TRAF2 and TRAF6 activation and the subsequent IκB degradation and NF-κB nuclear translocation. The intersection of Dectin-1 and TLR signaling may be at the step of TRAF6 whose activation by TLR2 is mediated through IRAK1.
