Podocytes were transfected with plasmids encoding different forms of RhoA (wt or consistent active mutant Q63L) or mDia2 (full-length mDia2 or mDia2ΔGBD). (A) Actin conformation in cells with different activity of RhoA or mDia2 was illustrated by phalloidin stain. Lamellipodia were illustrated by immunostain of cortactin. (B) Lamellipodial changes were quantified and statistically analyzed for 20 randomly selected cells in each group by measurement of cortical area or ruffle length with cortactin enrichment, as in Figure 1.
