Short term TCR stimulation stabilizes ThPOK in human CD4+ T cells. A–C, CD4 SP and CD8 SP T cells were isolated from human peripheral blood mononuclear cells of healthy donors. B, total RNA was extracted, and the mRNA levels of TIP60 and ThPOK were measured by qRT-PCR analysis. Data show the mean of three separate experiments. Error bars indicate S.D. C, cells were lysed with RIPA buffer; TIP60 and ThPOK protein levels were analyzed by Western blotting (IB) using β-actin as a loading control. D and E, resting CD4+ T cells were stimulated with α-CD3 and α-CD28 antibodies for the indicated time periods.
