Short term TCR stimulation stabilizes ThPOK in human CD4+ T cells.
A–C, CD4 SP and CD8 SP T cells were isolated from human peripheral blood mononuclear cells of healthy donors. B, total RNA was extracted, and the mRNA levels of TIP60 and ThPOK were measured by qRT-PCR analysis. Data show the mean of three separate experiments. Error bars indicate S.D. C, cells were lysed with RIPA buffer; TIP60 and ThPOK protein levels were analyzed by Western blotting (IB) using β-actin as a loading control. D and E, resting CD4+ T cells were stimulated with α-CD3 and α-CD28 antibodies for the indicated time periods. D, cells were lysed with RIPA buffer; cell lysates were directly tested by Western blotting with the indicated antibodies. E, total RNA was extracted, and the mRNA levels of ThPOK, TIP60, Eomes, and Tbx21 were measured by qRT-PCR. Data show the mean of at least three independent experiments. Error bars indicate S.D.
