Classical calpains, which include CAPN1, 2, 3b, 8, 9 11, 12, 13, 14, consists of two protease core domains (PC1 and PC2), a C2 like domain (C2L) and a penta EF-hand domain (PEF). The PC1 and PC2 make up the cysteine protease core. All calpains have a protease core domain, but the other domains differ in arrangement for the non-classical calpains.
