A) Pretreatment of neutrophils with Msp increases lipid phosphatase activity measured in partially permeabilized cells using a malachite green assay. B) Treatment of neutrophils with the bisperoxovanadate compound VO-OHpic, a PTEN inhibitor, prevented Msp-mediated phosphate release. C) Immunoprecipitated PTEN from cell lysates was used to measure specific PTEN activity following Msp pretreatment prior to fMLP stimulation. For these assays, activity was measured by incubation with soluble diC8 PIP3, and phosphate release was measured using the malachite green assay. Graphs represent the mean ± SEM of 3 independent experiments (* P<0.05, ** P<0.01, *** P<0.001 unpaired t test).
