Effects of EGCG on the expression levels and enzymatic activity of IDO in SW837 cells. Cells were pretreated with various concentrations (0, 10, 50 or 100 μM) of EGCG for 2 h and were then stimulated with IFN-γ for 24 h. (A) Total RNA was extracted and the IDO mRNA expression levels were measured by quantitative real-time RT-PCR. (B) Total cellular protein was extracted and the levels of IDO protein expression were determined by western blot analysis. (C) Functional IDO enzymatic activity was determined by measuring the concentrations of L-kynurenine in culture medium under the same conditions. Columns and bars indicate the mean and SD. Repeated western blots gave similar results.
